Abstract

The Trichoderma genus are widely used as biological control agents against various plant pathogens, however, many molecules involved in biocontrol are still unknown. There are numerous works in which the molecules secreted by the fungus are evaluated, among which are proteins. To evaluate these molecules, they must be extracted from the system in which the fungus grows, and the extraction methodology, which is highly dependent on various factors, requires adjustment of experimental conditions. Therefore, the aim was the evaluation of two methodologies for the analysis of the profile of secreted proteins by isolates of Trichoderma spp to the culture medium, starting using filtrate without lyophilization and filtrate lyophilizate. Protein profiles obtained from lyophilized material , applying protein precipitation with trichloacetic acid in acetone followed by cleaning with methanol and chloroform, generated an adequate profile for the analysis of proteins secreted into the culture medium.
